摘要: 以2个主栽枣树品种‘长红枣’和‘金丝小枣’为材料,研究了基因型、基本培养基、植物生长调节剂及碳源对枣花药愈伤组织发生及不定芽分化的影响。结果表明:‘长红枣’较‘金丝小枣’的离体花药更容易诱导出愈伤组织。适合‘长红枣’花药愈伤组织诱导的生长调节剂组合为1.0mg/L 2,4-D+2.0mg/L 6-BA和0.5mg/L NAA+2.0mg/L 6-BA,出愈率均在94.00%以上。基本培养基MS与1/2MS对‘长红枣’花药愈伤组织诱导影响差异不显著,使用麦芽糖较蔗糖更适宜诱导花药愈伤组织。在MS+6-BA 1.0~2.0 mg/L+NAA 0.5mg/L培养基中,‘长红枣’花药愈伤组织的出芽率超过60.00%。‘金丝小枣’愈伤组织诱导和出芽培养基分别为1/2MS+1.0mg/L 2,4-D和MS+6-BA 1.0mg/L+NAA 0.5mg/L,花药出愈率和愈伤出芽率分别达到56.17%和20.36%。
英文摘要: The effects of genotype,basic medium,plant growth regulator and carbon source on callus induction from in vitro anther and adventitious bud formation were investigated using two major cultivars of Chinese jujube(Ziziphus jujuba Mill.),‘Changhongzao'and‘Jinsixiaozao'.The results showed that the callus induction from anther was much easier in‘Changhongzao'than in‘Jinsixiaozao'.1.0mg/L 2,4-D + 2.0mg/L 6-BA and 0.5mg/L NAA +2.0mg/L 6-BA were appropriate plant growth regulator combinations for the anther callus induction in‘Changhongzao',in which the callus rate reached more than 94.00%.There was no significant difference of callus induction between MS and 1/2MS basic medium in‘Changhongzao'.The callus induction from anther was easier in medium supplemented with maltose than with sucrose.Adventitious bud rate of‘Changhongzao'was more than 60.00%in medium MS + 6-BA 1.0~2.0mg/L + NAA 0.5mg/L.In case of‘Jinsixiaozao',the callus ratereached 56.17%in medium 1/2MS+1.0mg/L 2,4-D and the adventitious bud rate was 20.36%in medium MS+6-BA 1.0mg/L + NAA 0.5mg/L.
