摘要: 以‘长富2号’(Nagafu No.2)红富士苹果试管苗为试材,研究了BA和TDZ组合、不同浓度的TDZ、糖的种类和不同暗培养时间对红富士苹果离体叶片再生不定芽的影响。结果表明:高浓度的TDZ可以有效提高红富士苹果离体叶片再生效率;最适宜叶片再生的糖类为蔗糖,最适宜的暗培养时间为3周,最佳的叶片再生培养基为MS+TDZ 6.0 mg/L+NAA 0.1 mg/L+蔗糖35 g/L+琼脂6.2 g/L。在上述培养条件下,‘长富2号’红富士苹果离体叶片再生率达到100%,平均叶片再生芽数达到10.6个。
英文摘要: Experiments were conducted with leaves collected from the test-tube plantlet of Fuji 'Nagafu No.2' to study the effects of different concentrations of TDZ,combination of BA and TDZ,dark incubation and glucide on adventitious shoot regeneration in vitro.The results showed that the leaf regeneration efficiency of Red Fuji was improved effectively with high concentration of TDZ.The optimum glucide was sucrose,the optimum dark incubation time was 3 weeks,and the optimum medium was MS+TDZ 6.0 mg/L+NAA 0.1 mg/L+sucrose 35 g/L +agar 6.2 g/L.With the culture conditions above,the leaf regeneration frequency of Red Fuji reached 100%,and the number of buds per leaf rose to 10.6.
