摘要: 为筛选具有抗热应激功效的理想中药组方,本试验依据清热解暑、化湿醒脾、安神通乳、理气消食的原则,从香薷、黄芩、麦冬、薄荷、木通、通草、藿香、茵陈、石菖蒲、金银花、酸枣仁、山楂、厚朴、陈皮、甘草等备选中药中选取不同药物组拟了3个不同组方,通过人工制造小鼠热应激模型,并用上述不同中药组方给小鼠灌服,比较用药组与对照组小鼠临床症状及与热应激相关的生理、生化指标的变化,进而筛选出抗热应激的理想组方.结果显示:三个组方均不同程度改善了小鼠的临床症状和血液生理生化指标,其中以"抗暑一号"的中剂量水平效果最佳:热应激情况下,阳性对照组中的K+,Cl-,AST,ALT浓度较阴性对照组显著升高(P<0.01),而Na+,TP, ALB,AKP浓度较阴性对照组显著降低(P<0.01)."抗暑一号"中剂量水平能有效改善热应激小鼠的临床症状和血液生理生化指标异常,与阳性对照组相比,差异均极显著(P<0.01),其中Na+,K+,TP,ALT浓度可回升至正常水平.为下一步临床用于奶牛热应激的防治提供了理论依据.为研究"抗暑一号"抗奶牛热应激的效果及其机理,本课题选取5头体重、年龄、胎次、产奶量相近的健康荷斯坦奶牛进行试验,临床试验结果表明:热应激阶段奶牛的平均呼吸频率、平均直肠温度均较常温阶段显著升高(P<0.01),而平均日单产奶量、乳脂率、乳蛋白率却显著降低(P<0.01).用药后奶牛平均呼吸频率、平均直肠温度比热应激阶段均有所下降,但差异不显著(P>0.05);而日单产奶量、乳脂率、乳蛋白率均显著回升(P<0.01).热应激情况下,奶牛血清中的Na+浓度较阴性对照组有所降低(P>0.05),而K+和CL-浓度均较阴性对照组均有所升高,其中,K+差异不显著(P>0.05),CL-差异极显著(P<0.01).应用"抗暑一号"后,Na+浓度均较阳性对照组有所回升(P>0.05);K+浓度均较阳性对照组有所降低(P>0.05);CL-浓度较阳性对照组有显著降低(P<0.01).热应激情况下,奶牛血清中的TP、G浓度均较阴性对照组呈降低趋势(P<0.05);ALB浓度较阴性对照组有所降低(P>0.05).应用"抗暑一号"后,TP、G浓度均较阳性对照组呈回升趋势,并与阴性对照组相比,差异显著(P<0.05);ALB浓度均较阳性对照组呈回升趋势,与阴性对照组相比,差异不显著(P>0.05).热应激情况下,奶牛血清中ALT、AST浓度较阴性对照组呈上升趋势,ALT差异不显著(P>0.05),AST差异极显著(P<0.01);血清中AKP浓度较阴性对照组呈降低趋势(P>0.05).应用中药"抗暑一号"后,奶牛血清中ALT、AST浓度较阳性对照组呈降低趋势,ALT差异不显著(P>0.05),AST差异极显著(P<0.01);血清中AKP浓度较阳性对照组呈上升趋势(P>0.05).热应激情况下,奶牛血清中的HSP70浓度较阴性对照组极显著升高(P<0.01),血浆中ALD浓度较阴性对照组极显著降低(P<0.01),血清中的COR、T4浓度极显著降低(P<0.01),ACTH显著降低(P<0.05),T3有所降低(P>0.05).应用中药方剂"抗暑一号"后,HSP70浓度较阳性对照组有所上升(P>0.05),血浆中ALD浓度较阳性对照组极显著升高(P<0.01),血清中的COR、T4浓度极显著升高(P<0.01),ACTH浓度显著升高(P<0.05),T3有所升高(P>0.05).由此表明中药组方在一定程度上能够预防和缓解奶牛热应激.
英文摘要: In order to selected the anti-heat stress traditional chinese medicine compounds on dairy cows,In this study, on the basis of clearing away summer heat and heat evil, dissipating dampness, calm the nerves, regulating vital energy, promoting digestion, the three different chinese herbal medicine prescriptions were organized with elsholtzia splendens,baikal skullcap root, Ophiopogon japonicus, mint, Akabia Stem, Rice Paper Plant Pith, Wrinkled Gianthyssop Herb, Capillary Wormwood Herb, Tatarinow Sweetflag Rhizome, honeysuckle, Spine Date Seed, hawthorn, Magnolia officinalis,Citrus,licorice, etc, Manufactured by artificial heat stress model in mice,and different Chinese Traditional Medicine decoction were given to the mice, compared with heat stress-related clinical symptomsphysiol and ogical and biochemical indexes between the treatment group and the control group, Selected the ideal prevention and treatment prescriptions on heat stress, By its application to test dairy cows, the results Proved that, to a certain extent, can prevent and mitigate on dairy cows under heat stress.The experimental results of screening Prescription showed that: All the three Prescription could improve the clinical symptomsphysiol and physiological and biochemical indexes of blood in heat stress mice. Among them, the middle dose Play the best effect :under the heat stress, the K+,Cl-,AST,ALT concentration in positive control group were increased significantly than those in negative control group(P<0.01), but Na+,TP, ALB,AKP concentration in positive control group were decreased significantly than those in negative control group(P<0.01).The middle dose of"Kangshuyihao"could improve the physiological and biochemical indexes of blood in heat stress mice, compared with the mice in positive control group, the effects were significant(P<0.01). Na+, K+,TP,ALT concentration could restore to the normal level.In order to study the effect and mechanism of"anti-summerⅠ"to heat stress, the subject selected five weight, age, parity, milk yield similar to the health of Holstein cows to test, the clinical trial results showed that:under the heat stress, the average respiratory rate and rectal temperature of dairy cows were significantly higher than room temperature phase (P <0.01), But the average daily milk yield, milk fat, milk protein rate was significantly lower (P <0.01).Cows after treatment, the average respiratory rate and rectal temperature were higher than the average stage of heat stress are decreased, but the difference was not significant (P> 0.05),while the milk yield,milk fat,milk protein were significantly recovered (P <0.01).Under the heat stress,the dairy cows sera Na+ concentration decreased than the negative control group (P> 0.05), and the K+ and Cl- concentration increased than the negative control group, Among them, K+ difference was not significant (P > 0.05), Cl--significant difference (P <0.01).After usage of traditional Chinese medicine prescription"Kangshu I", Na+ concentration recovered than the positive control group (P> 0.05). K+ concentration decreased than the positive control group (P> 0.05); Cl- concentration was significantly lower than the positive control group (P <0.01).Under the heat stress, dairy cow sera TP,G concentration decreased than the negative control group (P<0.05); ALB concentration decreased than the negative control group (P> 0.05). After usage of traditional Chinese medicine prescription"Kangshu I",TP,G concentrations showed a rising trend than the positive group, and compare with the negative control group, was significant difference (P<0.05); ALB concentration showed a rising trend than the positive control group, but compare with the negative control group, the difference was not significant (P>0.05).Under the heat stress,the dairy cows sera ALT,AST concentration in positive control group showed an upward trend than the negative control group,ALT was not significant (P> 0.05), AST was significantly different (P <0.01);sera AKP concentration was decreas (P> 0.05). After usage of traditional Chinese medicine prescription"Kangshu I", the dairy cows sera ALT,AST concentration in positive control group showed a decreasing trend,ALT was not significant (P> 0.05),AST was significantly different (P <0.01);serum AKP concentration showed an upward trend (P> 0.05).Under the heat stress,the dairy cows sera HSP70 concentration in positive control group increased significantly than those in negative control group(P<0.01);The plasma ALD concentration, sera COR and T4 concentrations decreased significantly than those in negative control group (P <0.01); ACTH decreased significantly than those in negative control group (P<0.05), T3 decreased non-significantly than those in negative control group (P>0.05).
