摘要: 为建立良好的棉花遗传转化系统,本研究采用基因枪轰击棉花成熟种子胚尖的方法,将本实验室克隆的抗黄萎病相关基因GhDAHPS转化到棉花中.抗生素抗性鉴定结果表明:在基因枪轰击的309个胚尖中,有90株为卡那抗性植株,进一步通过PCR检测这些抗性植株,其中有5株呈阳性,PCR阳性率为1.6%.结果表明:基因枪转化棉花成熟种子胚尖是可行的,相对于其他转化方法,具有操作简便,植株成苗较快,转化周期短等优点.
英文摘要: In order to establish a new cotton genetic transformation system,GhDAHPS gene from cotton,which was demonstrated to be related with verticillium wilt resistance,was tried to introduce into cotton via particle bombardment using embryonic tip explants.The results showed that 90 out of 309 embryonic tips were obtained after kanamycin selection culture,these resistant plants was further tested by PCR analysis,5 out of 90 transformed plants were PCR positive,PCR positive rate was 1.6%.This study showed that the ...
