摘要: 建立一种ELISA法检测牛奶中青霉素G残留方法.以卵清蛋白为载体,连接青霉素G,合成完全抗原,免疫兔子,获得抗血清,然后以此抗血清为一抗,酶标羊抗兔抗体为二抗,摸索ELISA操作的工作条件.结果表明一抗稀释倍数为1∶3 200,二抗稀释倍数为1∶100 000为最佳工作条件.建立了检测标准曲线,回归方程为y=(2.422 8+1.692 3*x1.1474)/(35.267 1+x1.1474),相关系数为0.999 9.在此条件下可以检测牛乳中的青霉素G的限度为(1~128)ng/mL.经重复性试验表明该法重现性好,适用于检测牛乳中的青霉素G残留.
英文摘要: The penicillin G residues in milk of cow were here determined with ELISA.The synthesis of complete antigen of penicillin G was performed by coupling with ovalbumin and penicillin G.The antiserum of penicillin G was obtained by immunizing rabbit.The procedure of ELISA was studied with antiserum of penicillin G as primary antibody and goat anti-rabbit antibody coupling HRP as secondary antibody.The optimized procedure of ELISA was determined by primary antibody as dilution of 1:3 200 and secondary antibody as...
