摘要: 以同源四倍体大白菜为试材,进行了游离小孢子培养获得初级三体、及其细胞学依据研究,结果表明:①游离小孢子胚胎发生能力因供体基因型而异.其中9405品系产胚率达81胚/蕾.②小孢子植株中,三体株率为14.79%-22.41%.③该途径获得三体的细胞学依据是由于同源四倍体大白的花粉母细胞内Ⅰ、Ⅲ、Ⅳ型配对的同源染色体发生不规则分离,后其Ⅰ、Ⅱ部分染色体落后和向两极分配不均衡,致使后期Ⅱ型染色体数目变化于15-24的各类细胞.具有21条染色体的小孢子(2x+1)再生植株能力与正常小孢子(2x)再生植株能力相近.④减数分裂的各种异常频率和三体株获得率品系间存在差异.
英文摘要: The primary trisomics from isolated microspore culture of autotetraploid Chinese cabbage and its cytological basis were studied. The results showed that① Embryogenesis were genotype dependent. The Line 9405 produced 81 embryos per bud which was the highest yield. ②The number of trisomics were 14.79%-22.41%among the microspore plants.③PMC chromosome pairing forms at diaknesis were Ⅰ , Ⅱ ,Ⅲ, Ⅳ. The irregular separation of Ⅰ,Ⅲ ,Ⅳ , straggling chromosomes at metaphase I resulted in the chromosome number at each pole of PMC at anaphase Ⅱ were varied from 15-24. The plant regeneration rate of microspore which had 21 chromosomes was similar to the normal microspore which had 20 chromosomes. ④The frequency of chromosome pairing forms at diakinesis and irregular behavior at metaphase Ⅰor Ⅱ, anaphaseⅠorⅡ were different among the strains 9402,9405,9409.
