摘要: 本试验研究发现野生型拟南芥Col-0生态型对核盘菌敏感,接菌6 h后,叶片上有明显的水渍状病斑且在接菌1h H2O2的产量最大,随着时间的延长H2O2产量逐渐减少,到6h已基本消失,利用含1.2 mmol/L草酸(OA)的MS培养基,对5000余株拟南芥T-DNA插入突变体进行初筛,获得126株突变体,将培养皿中筛选的具有OA抗性的突变体,进行接菌鉴定,筛选到1株抗病突变体.通过TAILPCR发现,T-DNA插入到锌指结构家族蛋白基因的3'非编码区上.
英文摘要: Wild-type Arabidopsis thaliana Columbia is susceptive to Sclerotinia sclerotiorum,hygrophanous spot can be found on the leaves treated S.sclerotiorum in 6 hours and the production of H2O2 reached maximum at 1 hour,then gradually decreased and disappeared to 6 hours.We screened 126 Arabidopsis mutants capable of surviving on MS agar medium with 1.2 mmol/L oxalic acid among 5 000 Arabidopsis mutants.At last,2 Arabidopsis mutants were screened with significantly smaller size of lesions.T-DNA inserted 3' untranslated region of zinc finger family protein by TAILPCR.
