摘要: 本试验旨在利用环境控制舱模拟我国冬季鸡舍内部环境状况来探究二氧化碳浓度对60周龄大午金凤蛋鸡生产性能、血气指标、血液生化指标及肺脏组织内皮素基因表达量等影响。选取健康、体重相近的60周龄“大午金凤”蛋鸡180只,随机分成3组,每组5个重复,每个重复12只鸡。Ⅰ组为对照组保持二氧化碳浓度为1 500mg/m~3不变,Ⅱ、Ⅲ组为试验组二氧化碳浓度分别为3 500mg/m~3和5 500mg/m~3,3个组的温度保持15℃不变。预试期1周,正式期8周。研究结果表明:(1)二氧化碳浓度对蛋鸡生产性能的影响主要体现在采食量、产蛋率和料蛋比上,且长时间处在5 500mg/m~3浓度下影响程度大。试验进行到第4周末,与对照组相比,5 500mg/m~3浓度组的采食量增加,料蛋比升高,且变化显著(P<0.05);第5-8周内与对照组相比,二氧化碳浓度为3 500mg/m~3和5 500mg/m~3试验组的采食量增加(P<0.05),产蛋率下降,5 500mg/m~3的试验组变化明显(P<0.01),料蛋比也显著高于对照组(P<0.01)。(2)二氧化碳浓度对营养物质代谢的影响较小,经测量发现在实验的第4周周末,二氧化碳对蛋鸡的钙和磷的物质代谢率影响较大,处于5 500mg/m~3二氧化碳浓度蛋鸡的钙和磷的物质代谢率较对照组显著下降(P<0.05)。第5-8周3 500mg/m~3组和5 500mg/m~3组与对照组相比,粗蛋白物质代谢率显著降低(P<0.05),且5 500mg/m~3组降低极显著(P<0.01);在整个过程能量的物质代谢率呈降低趋势(P>0.05);脂肪在第8周3 500mg/m~3组和5 500mg/m~3组与对照组相比显著降低(P<0.01),钙和磷的物质代谢化率也显著降低(P<0.05)。(3)二氧化碳浓度对蛋壳厚度的影响比较大,从第3周开始二氧化碳浓度3500mg/m~3组和5 500mg/m~3组与对照组相比蛋壳厚度有降低趋势(P>0.05),第4周开始5 500mg/m~3试验组较对照组的蛋壳厚度出现显著降低(P<0.05),随着时间的延长蛋壳厚度降低更为显著(P<0.01)。3 500mg/m~3试验组较对照相比蛋壳厚度在第6周后显著降低(P<0.05),随着时间延长降低趋势也更加明显。(4)二氧化碳浓度能够影响机体的血气指标,第4周3 500mg/m~3和5 500mg/m~3试验组较对照组血液PH显著降低(P<0.05);血液中的二氧化碳分压也显著升高(P<0.05);同时HCO_3~-浓度也显著升高(P<0.05)。第8周,5 500mg/m~3组较对照组相比二氧化碳分压和HCO_3~-浓度显著升高(P<0.05),3 500mg/m~3组有升高趋势但不显著(P>0.05)。(5)二氧化碳浓度能影响血清免疫球蛋白和补体含量,在第4周3 500mg/m~3和5 500mg/m~3试验组血液中的IgA、IgM、IgG较对照组出现显著降低(P<0.05)。补体C3、C4和白介素IL4、IL10在前4周变化不显著。试验后4周及6周和8周血液中免疫指标呈降低趋势,其中,3 500mg/m~3和5 500mg/m~3试验组血液中的IgA、IgM、IgG较对照组出现显著降低(P<0.05),补体C3、C4较对照组相比也显著降低(P<0.05)。(6)二氧化碳浓度对肺脏组织中内皮素相关基因表达有较大影响,第4周,5500mg/m~3试验组的ET-1和ET-A较对照组和3 500mg/m~3试验组的表达量显著升高(P<0.05),而ET-B的表达量有升高趋势但不显著(P>0.05),3 500mg/m~3试验组较对照组3个基因的表达量都有升高但不显著(P>0.05)。第8周,ET-1和ET-A基因的表达量与第4周的变化规律相同,5 500mg/m~3试验组受体ET-B较对照组和3500mg/m~3试验组的表达量显著升高(P<0.01)。(7)二氧化碳浓度对气管中相关基因表达有较大影响,各试验组的紧密连接蛋白claudin1、气管黏蛋白muc2呈现降低趋势,半胱天冬酶caspase3呈升高趋势。第4周,3 500mg/m~3和5 500mg/m~3试验组claudin1和muc2的表达量较对照组相比显著降低(P<0.01)。3500mg/m~3和5 500mg/m~3试验组caspase3的表达量较对照组呈升高趋势(P>0.05)。第8周,3 500mg/m~3和5 500mg/m~3试验组claudin1和muc2的表达量较对照组相比显著降低(P<0.01)。3 500mg/m~3和5 500mg/m~3试验组caspase3的表达量较对照组显著升高(P<0.01)。上述试验结果表明,二氧化碳浓度影响蛋鸡的生产性能,营养物质代谢、血液免疫指标和肺脏气管粘膜等相关基因的表达,高浓度的二氧化碳下,时间越久,对鸡的影响越大。
英文摘要: The objective of this study was to examine the effects of different carbon dioxide concentrationson production performance,blood gas index,blood biochemical indicators and lung tissue endothelin gene expression of laying hens in the internal environment of the chicken house in winter of China.18060thweek of age"Dawu Jinfeng"layers were selected,randomly divided into 3 groups,each group of 5replicates,replicate of 12 birds.Group I kept the carbon dioxide concentration at 1 500 mg/m~3 for the control group,Group II and III had the carbon dioxide concentration of 3500 mg/m~3 and 5 500 mg/m~3for the experimental group,and the temperature of the three groups remained at 15??C.The pre-test period is 1 week and the official period is 8 weeks.Research indicates:(1)The effect of carbon dioxide concentration on production performance of laying hens was mainly reflected in feed intake,egg production rate,and feed-egg ratio,and a long time under the influence 5500mg/m~3 concentration for a long time had a larger impact.At the 4th weekend of the trial,compared with the control group,the feed intake and the feed-egg ratio of the 5500 mg/m~3concentration group was increased,and the change was significant(P<0.05).Within 5-8 weeks,compared with the control group,the carbon dioxide concentration of 3500 mg/m~3 and 5 500 mg/m~3 of the experimental group the feed intake was increased(P<0.05),the egg production rate was decreased.The 5500 mg/m~3 group had significant changes(P<0.01),and the feed-egg ratio was also significantly higher than the control group(P<0.01).(2)Carbon dioxide concentration has little effect on nutrient metabolism.It was found that on the weekend of the fourth week of experiment,the apparent digestibility of calcium and phosphorus in laying hens was greatly influenced by carbon dioxide,and the apparent digestibility of calcium and phosphorus in laying hens at 5 500 mg/m~3 was significantly decreased than in the control group(P<0.05).At weeks 5-8,the apparent digestibility of crude protein in the 500 mg/m~3 and 5 500 mg/m~3groups was significantly lower than that in the control group(P<0.05),and significantly decreased in the 5 500 mg/m~3 group(P<0.01)£?The apparent digestibility of energy showed a decreasing trend in the whole process(P>0.05).At weeks 8,crude fat was significantly decreased in the 3 500 mg/m~3 group and5 500 mg/m~3 group compared with the control group(P<0.01).The apparent digestibility of calcium and phosphorus also decreased significantly(P<0.05).(3)Carbon dioxide concentration has a greater impact on egg shell thickness.Compared with the control group,the thickness of eggshells in the 3500mg/m~3 and 5500mg/m~3 groups began to decrease from the 3rd week(P>0.05).The thickness of egg shells in the 5 500 mg/m~3 trial group was significantly lower than that in the control group at the beginning of the 4th week(P<0.05),and the eggshell thickness decreased more significantly with time(P<0.01).The eggshell thickness of the 3 500 mg/m~3test group was significantly lower than that of the control group after the 6th week(P<0.05),and the trend was more pronounced as time went on.(4)Carbon dioxide concentration can affect the body's blood gas index.At the 4th week,the blood levels of PH of 3 500 mg/m~3 and 5 500 mg/m~3 test group were significantly lower than those of the control group(P<0.05)£?The partial pressure of carbon dioxide in blood was also significantly increased(P<0.05).At the same time,the HCO_3~-concentration was also significantly increased(P<0.05).At 8th week,5500mg/m~3 group had significantly higher carbon dioxide partial pressure and HCO_3~-concentration(P<0.05)than the control group,but the 3500mg/m~3 group had an increasing trend but not significant(P>0.05).(5)Carbon dioxide concentration can affect serum immunoglobulin and complement levels.In the4th week,IgA,IgM,and IgG in the blood of the 3 500 mg/m~3 and 5 500 mg/m~3 test groups were significantly lower than those in the control group(P<0.05).Complement C3,C4 and interleukin IL4,IL10 did not change significantly in the first 4 weeks.At 4 weeks and 6 weeks and 8 weeks,the immune index in the blood showed a decreasing trend.Among them,IgA,IgM,and IgG in the blood of the 3500mg/m~3 and 5500 mg/m~3 test group were significantly lower than those in the control group(P<0.05).Complement C3 and C4 were also significantly lower than those in the control group(P<0.05).(6)Carbon dioxide concentration has a great influence on the expression of endothelin-related genes in lung tissues.At the 4th week,the expression levels of ET-1 and ET-A in the 5 500 mg/m~3 test group were significantly higher than those in the control group and 3 500 mg/m~3 test group(P<0.05),while the expression level of ET-B was increasedbut not significant(P>0.05).The expression levels of ET-1,ET-A,ET-B genes in the 3 500 mg/m~3 test group were higher than those in the
