摘要: 为了评价板青颗粒的安全性,进行了小鼠的急性毒性试验与大鼠的亚慢性毒性试验,采用简化机率单位法测得小鼠最大耐受量(MTD)高于180 g/(kg·d),半数致死量(LD50)无法确定。进行亚慢性试验时将SD大鼠分为4组,每组20只,空白对照组饮服纯净水,板青颗粒低、中、高剂量组分别给予3,6,12 g/(kg·d)的板青颗粒于饮水中,连续28 d,分别检测体重、脏器指数、血常规指标、血液生化指标和组织切片观察。结果表明:与空白对照组比较,板青颗粒显著提高大鼠的脾脏指数(P〈0.05)、中性粒细胞百分比(Gran%)、血细胞比容(HCT)、红细胞体积分布宽度(RDW)(P〈0.05),显著降低胸腺指数、淋巴细胞百分比(Lymph%)、细胞血红蛋白浓度(MCHC)以及总蛋白(TP)、葡萄糖(GLU)、尿素氮(BUN)含量(P〈0.05);心脏、肝脏、脾脏、肺脏、肾脏组织切片无显著性病理变化。说明板青颗粒毒副作用和不良反应小,是安全性较高的中药方剂。
英文摘要: To evaluate the safety of Banqing granules, the acute and the subchronic toxicity tests were carried out in mice and SD rats, respectively. The maximum tolerant dose (MTD) of mice was detected by simplified probit method, which was more than 180 g/( kg· d), but the LDs0 could not be determined. SD rats were randomly divided into/bur groups of 20 rats each in the subchrenic toxicity test, including a blank control group which was given pure water, and the low - , medium - , high - dose treatment groups which were given 3,6, 12 g/( kg · d) of Banqing granules in drinking water for continuous 28 d. The body weight, organ indexes, routine blood test, blood biochemical indexes, were all detected, and the tissue sections were observed, respectively. The results showed that compared with the blank control group, Banqing granules significantly increased ( P 〈 0.05 ) the spleen index, the percentage of neutrophils ( Gran% ) , hematocrit (HCT) , and red blood cell distribution width ( RDW), and significantly decreased ( P 〈 0. 05 ) the thymus index, percentage of lymphocytes ( Lymph% ), the contents of mean corpuscular hemoglobin concentration (MCHC), total protein (TP), glucose (GLU), blood urea nitrogen (BUN). There were no significant pathological changes in the tissue sections from the heart, liver, spleen, lung, and kidney. The results indicate that the toxic side effects and adverse reactions of Banqing granules are samll, showing Banqing granules are safe Chinese medicinal formulae.
