摘要: 根据NCBI上公布的14个TaCDPK基因序列分别设计特异引物,将小麦品种‘洛夫林10’分别与叶锈菌生理小种260和165组成不亲和组合与亲和组合,在接菌后不同时间点分别取样,利用实时定量PCR技术检测14个TaCDPK基因在小麦与叶锈菌互作过程中的表达谱。结果显示,14个TaCDPK基因的表达模式可分为4类:TaCPK1、TaCPK2、TaCPK6、TaCPK7在叶锈菌侵染早期2个组合中的表达量均明显升高;TaCPK3、TaCPK18、TaCPK19在叶锈菌侵染早期2个组合中的表达量均明显下降;而TaCPK5、TaCPK8、TaCPK9、TaCPK12、TaCPK13、TaCPK15的表达量在不同组合中随接种时间的延长变化不明显;另外,TaCPK4在2种组合中前期表达量并无明显差异,但是接种后48h在不亲和组合中的表达量明显升高,而亲和组合中的表达量基本没有变化。结果表明,TaCPK1、TaCPK2、TaCPK6、TaCPK7、TaCPK3、TaCPK18、TaCPK19、TaCPK4可能参与小麦抵抗叶锈菌侵染过程;而TaCPK5、TaCPK8、TaCPK9、TaCPK12、TaCPK13、TaCPK15在小麦抗叶锈菌侵染中可能不起作用。
英文摘要: In the study,specific primers were designed based on the sequences of 14 wheat TaCDPK genes published on NCBI.The wheat variety ‘Lovering 10'was composed into incompatible and compatible combinations with leaf rust races 260 and 165,respectively.The expression patterns of TaCDPKs were detected by qRT-PCR at different time points after the inoculation.The results showed that the expression patterns of 14 TaCDPK genes could be divided into four categories:the expressions of TaCPK1,TaCPK2,TaCPK6 and TaCPK7 increased in the early interaction stage in both incompatible and compatible combinations;the expressions of TaCPK3,TaCPK18 and TaCPK19 reduced in the early interaction stage in both incompatible and compatible combinations;the expressions of TaCPK5,TaCPK8,TaCPK9,TaCPK12,TaCPK13 and TaCPK15 changed little in both combinations;the expressions of TaCPK4 had little difference between incompatible and compatible combinations during the early time but increased obviously in the incompatible combintion after 48 hours.It concluded that TaCPK1,TaCPK2,TaCPK6,TaCPK7,TaCPK3,TaCPK18,TaCPK19andTaCPK4 may play important roles in the interaction of wheat and leaf rust.However,TaCPK5,TaCPK8,TaCPK9,TaCPK12,TaCPK13 and TaCPK15 may have no functions in the wheat-leaf rust interaction.
