摘要: 以紫叶挪威槭2年生枝条水培后展开1~2片小叶的萌发芽为试验材料,采用无菌条件下组织培养的方法,研究了紫叶挪威槭外植体材料不同的消毒方法和时间,不同的不定芽和不定根分化培养基对再生体系建立的影响,以期低污染、高效的建立紫叶挪威槭的再生体系。结果表明:萌发芽经2%洗涤灵溶液洗涤,无菌水冲洗,去除残留,70%乙醇处理30s,无菌水冲洗5~6次,1%次氯酸钠消毒液处理20min,无菌水冲洗5~6次,接种到不定芽分化培养基MS+0.05mg/L NAA+1.0mg/L 6-BA+0.3g/L CH(水解酪蛋白)+2.0g/L PVP+30g/L蔗糖(pH5.5)诱导并产生不定芽。不定芽继代至不定根分化培养基MS+0.4mg/L IBA+0.05mg/L NAA+0.3g/L CH+2.0g/L PVP+25g/L蔗糖(pH 5.5)诱导形成不定根,建立了紫叶挪威槭的植株再生体系。
英文摘要: Using the germination of the buds with 1-2pieces leaflet as test material,the influence of different disinfection methods,different time and different mediums with the method of tissue culture in aseptic conditions were studied,in order to establish plant regeneration system of Acer platanoides L.‘Crimson King'in low pollution and high efficiency.The results showed that the germination of the buds were treated with 2%detergent and flushed with sterile water.Then they were treated with 70%alcohol for 30 seconds and flushed with sterile water for 5-6times.After germination of the buds were treated with 1%sodium hypochlorite for 20 minutes and flushed with sterile water for 5-6times,they shoud be inoculated into MS+0.05mg/L NAA+1.0mg/L 6-BA+0.3g/L CH+2.0g/L PVP+30g/L sucrose(pH 5.5)medium.The plant regeneration system of Acer platanoides L.‘Crimson King'could be quickly established,when the plantlets were subcultured into MS+0.4mg/L IBA+0.05mg/L NAA+0.3g/L CH+2.0g/L PVP+25g/L sucrose(pH 5.5).
